CIP@Cornell Seminar: Manuel Leonetti, Chan Zuckerberg Biohub SF

Join the Center for Innovative Proteomics (CIP@Cornell) at Weill Institute for a seminar with special guest Dr. Manuel D. Leonetti, Ph.D. Dr. Leonetti is a Sr. Group Leader, Quantitative Cell Science Director, Systems Biology at Chan Zuckerberg Biohub SF where he is pioneering the breakdown of how human cells are built through his flagship project, OpenCell, at opencell.czbiohub.org. Talk Title: Intracellular Cartography: Cell Biology across Cellular Scales Abstract: Understanding how the proteome is spatially organized within cells remains a central challenge in cell biology. Here, I will present integrative methods for mapping the intracellular architecture of the human proteome at scale. We combine CRISPR-based endogenous tagging, fluorescence live-cell imaging, and quantitative mass spectrometry to generate high-resolution spatial and interaction proteomic maps of human cells. Using graph-based analysis and machine learning, we delineate dynamic protein networks, uncover functional microdomains, and systematically annotate subcellular localization. I will highlight recent advances from our OpenCell and organelle proteomics efforts, including a global map of subcellular organization derived from native immunoprecipitation of tagged organelles. Our work highlights that many cellular responses to perturbation are governed by spatial remodeling rather than by abundance changes. Together, these approaches build a quantitative foundation for charting the cell as a dynamic, spatially organized proteome. Bio: Manuel Leonetti has a long-term interest in mapping the architecture of biological systems. As a Ph.D. student with Rod MacKinnon at Rockefeller University, Leonetti used X-ray crystallography and electrophysiology to study the relationship between atomic structure and function of ion channel proteins. As a post-doc with Jonathan Weissman at UCSF, he developed high-throughput CRISPR methods to precisely and efficiently edit the human genome. At the CZ Biohub, Leonetti leads a multidisciplinary group of biologists, engineers, and data scientists that strive to understand how human cells are built. The group develops and deploys technologies to illuminate the wiring diagram of proteins within the cell using a combination of genome engineering, live-cell imaging and mass spectrometry. They seek to characterize fundamental mechanisms in physiology and disease, but also to “reverse engineer” the cell—to understand the details of how it is built in order to predictably tune its properties and behavior. One of their goals is to build open datasets, software, and protocols for the entire scientific community to use and explore. Check out their flagship project, OpenCell, at opencell.czbiohub.org. CIP@Cornell aims to establish a community that will include all proteomics labs and users on Cornell campuses, and a “town square” where we will regularly gather through seminars with both internal and external speakers.
